The point of the present investigation was to blend effective chitosan nanoparticles (CNPs) loaded with Pterocarpus marsupium (PM) heartwood extract and evaluate its biomedical applications. Various plant extract concentrations (PM-CNPs-1, PM-CNPs-2, PM-CNPs-3) are used to synthesize chitosan nanoparticles and optimized to acquire a stable nanoparticle formulation. The entrapment efficiency and in vitro release studies of the plant extract encapsulated in CNPs are estimated. Standard Dosage: 3-6 g heartwood powder twice daily; 50-100 mL decoction twice daily; Vijaysar tumbler water: 200 mL morning on empty stomach; 500 mg-1 g standardized extract capsule twice daily
Bioavailability: Pterostilbene (primary active stilbene) has remarkably good oral bioavailability (~80%) due to dimethylation of hydroxyl groups (compared to resveratrol’s ~20%), better metabolic stability, and enhanced lipophilicity. Marsupsin shows moderate oral bioavailability (~30-40%). Epicatechin (flavanol) has moderate bioavailability (~20-30%). The traditional ‘tumbler method’ (overnight water extraction in Vijaysar wood cup) provides slow extraction of water-soluble actives including epicatechin and kino-tannic acid.
Optimal Timing: Morning on empty stomach for maximum glycemic impact; 30 minutes before meals; tumbler water first thing in morning
Standardized Extract: Heartwood extract standardized to >5% pterostilbene and >2% epicatechin by HPLC; total polyphenols >20% (as tannic acid equivalent); marsupsin content >1%
Shelf Life: Heartwood chips: 36-48 months; Powder: 18-24 months; Capsules/Tablets: 24-36 months; Vijaysar tumbler: 6-12 months (replace when color fading stops); Aqueous extract: 12-18 months
Storage: Heartwood and powder in airtight containers at 15-25 deg C. Protected from moisture. Vijaysar tumblers should be kept dry when not in use. Standardized extracts in light-resistant packaging (pterostilbene is moderately photosensitive).
