# Pippali Moola

> Pippali Moola (Piper longum L. (root)) is a plant used in Ayurveda, from the Piperaceae family. Piper longum referenced in WHO Traditional Medicine Strategy documents; not specifically monographed by WHO; listed in Ayurvedic Pharmacopoeia of India; part of Trikatu (three pungents) classical formulation.

## Key facts

- **Botanical name:** Piper longum L. (root)
- **Family:** Piperaceae
- **Pharmacopoeia status:** Piper longum referenced in WHO Traditional Medicine Strategy documents; not specifically monographed by WHO; listed in Ayurvedic Pharmacopoeia of India; part of Trikatu (three pungents) classical formulation

## Names and identification

| Language | Name |
|----------|------|
| English | [Pippali](/herb/pippali/) Moola |
| Latin/Botanical | *Piper longum L. (root)* |

## Key Phytochemical Constituents

- [Piperine](/reference/piperine/)
- [Piperlongumine](/glossary/compounds-o-q/#piperlongumine)
- [Piperlonguminine](/glossary/compounds-o-q/#piperlonguminine)
- Pipermonaline
- Piperundecalidine
- Sylvatin
- [Sesamin](/glossary/compounds-q-t/#sesamin)
- Diaeudesmin
- Beta-[sitosterol](/glossary/compounds-q-t/#sitosterol)
- Cepharadione A

## How does it work?

- Piperine enhances bioavailability of co-administered drugs/nutrients by inhibiting CYP3A4 and P-glycoprotein efflux pump, and stimulating intestinal amino acid transporters; root has higher piperine concentration than fruit
- Piperlongumine selectively kills cancer cells by elevating reactive oxygen species (ROS) through inhibition of glutathione S-transferase pi 1 (GSTP1), sparing normal cells with adequate [antioxidant](/reference/antioxidant/) defenses
- Sesamin and diaeudesmin from root exhibit anti-inflammatory effects through dual inhibition of COX-2 and 5-LOX pathways, reducing both prostaglandin and leukotriene synthesis

## Which traditional uses are supported by research?

- Bioavailability enhancement (Yogavahi) validated - piperine's ability to increase absorption of co-administered compounds is well-established pharmacologically, confirming the traditional Ayurvedic concept of Yogavahi (bioenhancer)
- Respiratory disease treatment (Shwasa, Kasa) supported by documented anti-inflammatory, bronchodilatory, and [immunomodulatory](/reference/immunomodulatory/) effects, though high-quality RCTs are still needed
- Deepana-Pachana (digestive stimulant) activity validated through studies showing enhanced gastric secretion and gastrointestinal motility

## What do recent clinical trials show?

## Recent safety updates

- [Pippali](/herb/pippali/) Moola used in over 135 classical Ayurvedic formulations with long traditional safety record; however, no standardized human clinical safety data available
- Piperine's bioavailability enhancement can increase plasma levels of co-administered drugs (phenytoin, theophylline, propranolol); significant drug interaction potential requiring monitoring
- Avoid high doses during pregnancy (potential uterotonic effects); may exacerbate gastric acidity in patients with GERD or peptic ulcers

## Dosage forms and preparation

**Dosage Forms:** Churna (root powder), Kwatha (decoction), Capsule, Tablet, [Ghrita](/herb/ghrita/), Vati

**Standard Dosage:** 1-3 g root powder daily; 15-20 mL decoction twice daily; 500 mg standardized root extract twice daily

**Bioavailability:** Piper longum root contains piperine at lower concentration than fruit (1-2% vs 4-8%), but has higher piperlongumine content. Root [alkaloids](/glossary/compounds-1-a/#alkaloids) show moderate-to-good oral bioavailability (30-50%). Root-specific sesquiterpenes and [lignans](/glossary/compounds-g-l/#lignans) contribute to distinct pharmacological profile compared to fruit. Bioenhancer activity is present but less potent than fruit piperine due to lower concentration.

**Optimal Timing:** After meals for digestive benefits. Before meals with honey for respiratory conditions. Part of [Trikatu](/herb/trikatu/) or digestive formulations taken with first morsel of food.

**Standardized Extract:** [Pippali](/herb/pippali/)-Moola extract (6:1) standardized to piperine >2%, piperlongumine >1.5%, total alkaloids >4%. HPTLC fingerprint distinguishing root from fruit extract.

**Shelf Life:** Dried root: 18-24 months; Root powder: 18-24 months; Extract: 24 months; Capsules: 24-36 months

**Storage:** Dried root: cool, dry, ventilated storage, below 25 deg C, protected from pest infestation. Powder and extracts: airtight containers, below 25 deg C, RH <55%, protected from light.

**Marker Compounds:** Piperine, Piperlongumine, Piperlonguminine, Methyl piperate, Sesamin, Dihydrostigmasterol, [Piperanine](/glossary/compounds-o-q/#piperanine), Brachystamide B

### Extraction Methods
