## How does it work?

- Moringa isothiocyanate-1 (MIC-1) activates Nrf2 pathway by releasing Nrf2 from Keap-1 complex, increasing nuclear translocation and expression of antioxidant enzyme genes (NQO1, HO-1, GCLC).
- MIC-1 suppresses NF-kB pathway by preventing phosphorylation of IkB and nuclear translocation of NF-kB, reducing transcription of pro-inflammatory genes.
- Dual NF-kB suppression and Nrf2 activation provides simultaneous anti-inflammatory and antioxidant effects.
- MIC-1 upregulates glutathione S-transferases (GST), preventing cell damage through reduced glutathione reactions with xenobiotics and ROS.
- Suppresses pro-inflammatory cytokines in LPS-stimulated macrophages.
- Reduces reactive oxygen species in high-[glucose](/glossary/compounds-d-g/#glucose)-treated human renal proximal tubule cells (HK-2), relevant to diabetic nephropathy.
- Transcriptomic analysis shows MIC-1 modulates inflammation, immunity, and oxidative stress pathways simultaneously.

## Recent safety updates

- Avoid consumption of root and root extracts - these parts contain potentially toxic compounds.
- Safety and efficacy in pregnancy and lactation is not established - information lacking.
- May inhibit conversion of thyroxine (T4) to triiodothyronine (T3) in animal studies - potential interaction with levothyroxine.
- May decrease how quickly the liver breaks down some medications (CYP enzyme inhibition), potentially increasing drug effects and side effects.
- May lower blood sugar - caution with diabetes medications to avoid hypoglycemia.
- May lower blood pressure - potential additive effect with [antihypertensive](/glossary/pharmacology/#antihypertensive) drugs.
- Generally well tolerated at doses up to 4 g with no significant increase in adverse events in clinical studies.
- Transient diarrhea is the most common side effect.
- Rare case reports: Stevens-Johnson syndrome reported, and cutaneous toxicity with respiratory distress and tongue edema following moringa powder consumption.

## Dosage forms and preparation

**Standard Dosage:** 3-6 g leaf powder per day; 500-1500 mg standardized leaf extract daily; 5-10 mL Moringa oil for culinary/topical use; 1-2 g seed powder for water purification

**Bioavailability:** Isothiocyanates (from glucosinolates) are well absorbed (~70-80%). [Quercetin](/reference/quercetinquercetin/) [glycosides](/glossary/compounds-g-l/#glycosides) undergo intestinal hydrolysis with moderate aglycone absorption (~25%). Beta-carotene bioavailability is fat-dependent - co-administer with lipids. Iron and calcium in Moringa leaves show 20-30% absorption rate. Moringa oleifera coagulant protein shows local GI activity without significant systemic absorption.

**Optimal Timing:** Morning with breakfast for nutritional supplementation; before meals for glycemic management; leaf tea between meals; Moringa oil topically at night for skin health

**Standardized Extract:** Shade-dried leaf powder standardized to minimum 1% isothiocyanates, 5% protein, and 0.5% total flavonoids. Leaf extract (10:1 hydroalcoholic) standardized to minimum 3% quercetin equivalents. Cold-pressed seed oil (Ben oil) meeting cosmetic grade standards. Seed protein isolate for water purification applications.

**Shelf Life:** Dried leaf powder: 18-24 months. Capsules/tablets: 24 months. Ben oil: 36+ months (exceptional oxidative stability). Tea bags: 18 months. Fresh leaves: 3-5 days refrigerated.

**Storage:** Leaf powder in airtight, light-resistant containers below 25 deg C (beta-carotene and [chlorophyll](/glossary/compounds-c-d/#chlorophyll) are photosensitive). Ben oil at room temperature in dark glass bottles. Capsules in blister packs. Fresh leaves at 4-8 deg C. Protect all forms from humidity.
